If you see eggs and young larvae then the queen is there, not necessary to lay eyes on her. A queenless colony has no young brood and makes a peculiar sort of sad slow buzzing sound when you open the hive. If given a comb of brood from another hive (must have newly laid eggs) a queenless colony can raise a new queen.
